ABSTRACT

One major strategy to generate genetically modified mouse models is gene targeting in mouse embryonic stem (ES) cells, which is used to produce gene-targeted mice for wide applications in biomedicine. However, a major bottleneck in this approach is that the robustness of germline transmission of gene-targeted ES cells can be significantly reduced by their genetic and epigenetic instability after long-term culturing, which impairs the efficiency and robustness of mouse model generation. Recently, we have established a new type of pluripotent cells termed extended pluripotent stem (EPS) cells, which have superior developmental potency and robust germline competence compared to conventional mouse ES cells. In this study, we demonstrate that mouse EPS cells well maintain developmental potency and genetic stability after long-term passage. Based on gene targeting in mouse EPS cells, we established a new approach to directly and rapidly generate gene-targeted mouse models through tetraploid complementation, which could be accomplished in approximately 2 months. Importantly, using this approach, we successfully constructed mouse models in which the human interleukin 3 (IL3) or interleukin 6 (IL6) gene was knocked into its corresponding locus in the mouse genome. Our study demonstrates the feasibility of using mouse EPS cells to rapidly generate mouse models by gene targeting, which have great application potential in biomedical research.

ABSTRACT

Objective: To study laparoscopic surgery for ectopic pregnancy complicated by hemorrhagic shock clinical efficacy and nursing research. Methods: Retrospective analysis of 60 cases of ectopic pregnancy complicated by hemorrhagic shock patients, divided into:the experimental group and control group of 30 cases, control group using traditional open surgery, laparoscopic surgery treatment groupclosely observe the clinical efficacy of the two groups. Results: Laparoscopic treatment of ectopic pregnancy in patients with hemorrhagic shock complicating operative time, blood loss than the control group; laparoscopic treatment of ectopic pregnancy in patients with hemorrhagic shock complicated exhaust time, postoperative morbidity and hospital stay was significantly lower than control group. Conclusion: Ectopic pregnancy laparoscopic surgery can improve concurrency cure rate of patients with hemorrhagic shock, better retention in patients with reproductive function, improve the patient's quality of life.

ABSTRACT

Alkaloids and lignans from the stems of Piper betle were studied. Compounds were isolated and purified by repeated silica gel, reverse phase silica gel, Sephadex LH-20 column chromatography and preparative thin layer chromatography. The structures were elucidated on the basis of spectral analysis. From the ethyl acetate soluble fractions of the 70% acetone extract, ten compounds were isolated and identified as piperine (1), pellitorine (2), N-isobutyl-2E,4E-dodecadienamide (3), dehydropipernonaline (4), piperdardine (5), piperolein-B (6), guineensine (7), (2E,4E)-N-isobutyl-7-(3',4'-methylenedioxyphenyl)-2,4-heptadienamide (8), syringaresinol-O-beta-D-glucopyranoside (9),pinoresinol (10). All Compounds were isolated from the plant for the first time, and compounds 9 and 10 were isolated firstly from the genus.
